Tissue specific as well as ubiquitous
gene knock down can be performed in transgenic rabbits through
interference RNA techniques. Transgenic rabbits can be prepared
for single gene as
well as for multiple gene knock
down.
As
example, BioProtein Transgenics has prepared transgenic rabbit
lines under expressing 4 milk
protein genes simultaneously
with high efficiency. Extinction
of targeted milk protein genes varied from 6
to 98% depending on the transgenic lines obtained
and interference RNA sequences used.
The knock down strategy developed by
the Company allows simultaneous
extinction of multiple genes and expression
of transgenes in the mean time.

The
kappa casein was not targeted in this knock down experiment
and serves as control. WAP, alfaS1 casein, beta casein,
and transferrin expression levels measured in milk
was significantly lowered depending on transgenic
lines and iRNA used. Expression levels moved as shown
below:

GENE OVER-EXPRESSION

Classical
gene over-expression can be obtained trough pronucleus
embryo micro-injection. The company owns strong transgenesis
expertise both with plasmidic DNA,
and larger DNA fragments (BACs).
Co-expression of transgene can be performed both from
the co micro-injection of two monocistronic
transgenes or from the micro-injection
of bicistronic transgenes.
